Fusion of Large Unilamellar Liposomes Containing Hemocyanin with Planar Bilayer Membranes

چکیده
Membrane Fusion, Large Liposomes, Planar Bilayer, Hemocyanin Channel, Asymmetrical Insertion Large unilamellar vesicles were prepared by detergent removal from micelles containing phosphatidylcholine, phosphatidylethanolamine and phosphatidylserine. Liposomes were then interacted with Megathura crenulata hemocyanin, a well studied channel former. Incubation of the resulting proteoliposomes on one side of a phosphatidylserine-containing planar bilayer under fusion conditions yielded strong current increases. Such increase is due to insertion of ionic channels from the liposomes into the planar bilayer. Studying the effects of Ba2+ on the electrical properties of the channel we could show that the protein is always inserted into a bilayer during this process, i.e. fusion of proteoliposomes with the artificial m em brane occurs. The strong non linearity of the current-voltage curve of the hemocyanin pore could be used as a probe of the extent to which fusion preserves the orientation of the channel through the bilayer.

Parameters affecting the fusion of unilamellar phospholipid vesicles with planar bilayer membranes
It was previously shown (Cohen, F. S., J. Zimmerberg, and A. Finkelstein, 1980, J. Gen. Physiol., 75:251-270) that multilamellar phospholipid vesicles can fuse with decane-containing phospholipid bilayer membranes. An essential requirement for fusion was an osmotic gradient across the planar membrane, with the vesicle-containing (cis) side hyperosmotic with respect to the opposite (trans) side....
متن کاملPolyethylene Glycol Repairs Damaged Membrane; Biophysical Application of Artificial Planar Bilayer to Mimic Biological Membrane
Polyethylene glycol (PEG) is a hydrophilic polymer, known to be capable to fuse numerous single cells in vitro, to join the membranes of adjacent neurons and giant invertebrate axons, and to seal damaged neural membranes. The molecular mechanism of the action of PEG is still unknown. It is believed that PEG dehydrates membranes and enables their structural components to resolve and rearrange in...
متن کاملFusion of liposomes with mitochondrial inner membranes.
A procedure is outlined for the fusion of mixed phospholipid liposomes (small unilamellar vesicles) with the mitochondrial inner membrane, which enriches the membrane lipid bilayer 30-700% in a controlled fashion. Fusion was initiated by manipulation of the pH of a mixture of freshly sonicated liposomes and the functional inner membrane/matrix fraction of rat liver mitochondria. During the pH f...
متن کاملMembrane events involved in myoblast fusion
Myoblast fusion has been studied in cultures of chick embryonic muscle utilizing ultrastructural techniques. The multinucleated muscle cells (myotubes) are generated by the fusion of two plasma membranes from adjacent cells, apparently by forming a single bilayer that is particle-free in freeze-fracture replicas. This single bilayer subsequently collapses, and cytoplasmic continuity is establis...

The transport of sterols incorporated into the lecithin bilayer of small unilamellar liposomes through a model membrane was studied. A two-chamber diffusion cell containing liposomes with incorporated [4-14C)cholesterol or beta-[4-14C]sitosterol in the donor chamber and liposomes with unlabeled cholesterol in the receiver chamber was used. The permeability coefficients of the sterols through si...
